Generic Name: Agomelatine
Indications:
Major depressive disorder in adults, especially in patients not responding to or intolerant of selective serotonin reuptake inhibitors (SSRIs)
Prevention of relapse in major depressive disorder
Pharmacology:
Agomelatine is a melatonergic agonist (MT1 and MT2 receptors) and a 5-HT2C antagonist.
It helps to resynchronize circadian rhythms.
It increases noradrenaline and dopamine release specifically in the frontal cortex.
Dosage & Administration:
The effective dose is 25 mg per day, taken once at bedtime.
The dose may be increased to 50 mg per day in patients with an inadequate response after two weeks.
It can be taken with or without food.
Contraindications:
Hypersensitivity to the active substance or any of the excipients
Hepatic impairment (i.e., cirrhosis or active liver disease) or transaminases exceeding three times the upper limit of normal
Concomitant use of potent CYP1A2 inhibitors (e.g., fluvoxamine, ciprofloxacin)
Side Effects:

Headache
Nausea
Diarrhea
Increased liver enzyme levels
