Amit 10mg is a prescription medication primarily used as a tricyclic antidepressant, especially when a sedative effect is desired. It's also indicated for the management of nocturnal enuresis (bedwetting) in children,prophylaxis of migraine,tension headache, and various forms of chronic pain.

How Amit Works (Pharmacology)

Amit contains amitriptyline, a tricyclic antidepressant with notable anticholinergic and sedative properties. While its exact mechanism isn't fully understood, it's believed to work by inhibiting the re-uptake of neurotransmitters like norepinephrine (NE) and serotonin (5-HT) at nerve endings in the brain. This increases their concentration in the synaptic clefts, which is thought to help regulate mood and alleviate depressive symptoms.   • 7-10 years: 10-20 mg at night.
  • 11-16 years: 25-50 mg at night.
  • Maximum treatment period, including gradual withdrawal, is 3 months. A full physical examination is required before any further course.
• Prophylaxis of Migraine: 100 mg daily.
• Tension Headache: 10-25 mg three times daily.

Important Considerations & Warnings

Side Effects: Possible side effects include (but are not limited to):

• Anticholinergic effects: Dry mouth, blurred vision, excessive perspiration, hyperpyrexia (high fever), urinary retention, urinary tract dilatation.
• Cardiovascular reactions: Hypotension (low blood pressure), syncope (fainting), postural hypotension (dizziness upon standing), hypertension (high blood pressure), tachycardia (fast heart rate), palpitations, myocardial infarction.
• CNS and Neuromuscular: Confusional states, disturbed concentration, disorientation, delusions.
• Allergic reactions: Skin rash, urticaria (hives), photosensitization.
• Haematological: Bone-marrow depression.
• Gastrointestinal: Nausea, epigastric distress, vomiting, anorexia, stomatitis, unpleasant taste, weight loss, diarrhea, constipation.
• Endocrine: Testicular swelling, gynecomastia (breast enlargement in males), breast enlargement, galactorrhea (milk production).

Precautions & Warnings:

• Use with caution in patients with a history of epilepsy, glaucoma, urinary retention, cardiac disease, diabetes, hepatic impairment, thyroid disease, increased intra-ocular pressure, and psychoses (as it may aggravate mania).
• Initially, sedation may affect your ability to drive or operate machinery.

Pregnancy & Lactation:

• Pregnancy Category C: Amitriptyline crosses the placenta. It should be used during pregnancy only if the potential benefit to the mother justifies the potential risk to the fetus.
• Lactation: Amitriptyline is excreted into breast milk. A decision should be made whether to discontinue nursing or the drug, considering the importance of the drug to the mother, due to potential for serious adverse reactions in nursing infants.
