Indications:
Acute and chronic schizophrenic disorders, particularly those with positive symptoms (delusions, hallucinations, thought disorders) and/or negative symptoms (blunted affect, emotional and social withdrawal).
Dosage and Administration:
Adults:
For acute psychotic episodes, the recommended oral dose is 400 mg to 800 mg daily, divided into two doses.
The daily dose may be increased up to 1200 mg/day in individual cases. Doses above 800 mg/day are not shown to be more effective and may increase adverse events.
Dosage should be adjusted according to individual response.
For patients with mixed positive and negative symptoms, doses should be adjusted to achieve optimal control of positive symptoms.
Patients with predominantly negative symptoms: Oral doses of 50 mg to 300 mg daily are recommended. Dosage should be adjusted individually.
The medicine should be taken as advised by the doctor.
Contraindications:
Hypersensitivity to amisulpride or any other ingredients in the formulation.
Prolactin-dependent tumors (e.g., pituitary gland prolactinomas, breast cancer).
Pheochromocytoma.
Children up to puberty.
Pregnancy and lactation.
Combination with MAOIs.
Warnings and Precautions:
Neuroleptic Malignant Syndrome (NMS): A potentially fatal syndrome associated with antipsychotic drugs, including Aride. Discontinue all antipsychotics, including Aride, if NMS is suspected.
Seizures: Aride can lower the seizure threshold. Monitor patients with a history of seizures closely.
Withdrawal Symptoms: Involuntary movement disorders (akathisia, dystonia, dyskinesia) have been reported following abrupt cessation of high doses of antipsychotics. Gradual withdrawal is recommended.
Prolactin Levels: Aride may increase plasma prolactin levels, leading to galactorrhea, amenorrhea, gynecomastia, breast pain, orgasmic dysfunction, and impotence.
QT Interval Prolongation: Use with caution in patients with cardiovascular disease, a family history of QT prolongation, hypokalemia, and other risk factors for QT prolongation.
Elderly Patients: Use with caution due to the potential risk of hypotension or sedation.
Renal Impairment: Dose adjustment is recommended.
Liver Impairment:
Pregnancy: Aride should be used during pregnancy only if the potential benefits justify the potential risks.
Lactation: Aride is found in breast milk and is contraindicated during breastfeeding.
Others:
Increased mortality in elderly patients with dementia-related psychosis.
Potential for venous thromboembolism.
Leukopenia, neutropenia, and agranulocytosis have been reported.
Hyperglycemia and impaired glucose tolerance.
