Deflux Suspension - (5mg/5ml)

Presentation: Each 5 ml contains Domperidone Maleate BP equivalent to Domperidone 5 mg. Available in bottles of 100 ml.  

Indications: Deflux (Domperidone) is indicated for the treatment of:

  • Dyspeptic symptom complex, often associated with delayed gastric emptying, gastro-oesophageal reflux and oesophagitis:  
    • Epigastric sense of fullness, feeling of abdominal distension, upper abdominal pain.  
    • Eructation, flatulence,early satiety.  
    • Nausea and vomiting.  
    • Heartburn with or without regurgitation of gastric contents.   
  • Acute nausea and vomiting of functional, organic, infectious, dietetic origin or induced by radiotherapy or drug therapy or induced in migraine.  
  • Parkinson's disease: In dopamine-agonist induced nausea and vomiting.  

Dosage and Administration:

  • Should be taken 15-30 minutes before meals.  
  • Adults: 10-20 mg (1-2 teaspoonfuls) every 6-8 hours daily. Maximum dose is 80 mg per day.  
  • Children: 0.2-0.4 mg/kg (2-4 ml suspension per 10 kg body weight) every 6-8 hours daily.
  • In acute nausea and vomiting, the maximum period of treatment is 12 weeks.

Pharmacology: Domperidone is a peripheral dopamine-receptor blocker. It increases oesophageal peristalsis, enhances gastric emptying and duodenal peristalsis and regulates contraction of the pylorus. It has little effect on dopaminergic receptors in the brain, thus lacking psychotropic and neurologic side effects.

Side Effects: Side effects are uncommon. Occasionally, transient intestinal cramps may occur. Extrapyramidal reactions are rare in young children and exceptionally rare in adults. These symptoms resolve spontaneously and completely upon discontinuation of treatment. Domperidone may increase prolactin levels, potentially leading to galactorrhoea and gynaecomastia in rare cases. Dry mouth, thirst, headache, nervousness, drowsiness, diarrhoea, skin rash and itching have been reported rarely.

Precautions: Should be used with caution in young children due to a potentially increased risk of extrapyramidal reactions because of an incompletely developed blood-brain barrier. Use with caution in patients with hepatic impairment. In patients with severe renal insufficiency, the dosing interval should be increased to once or twice daily depending on the severity of the impairment.

Contraindications: Hypersensitivity to domperidone. Should not be used whenever gastrointestinal stimulation might be dangerous (e.g., gastrointestinal haemorrhage, mechanical obstruction or perforation). Contraindicated in patients with prolactin-releasing pituitary tumour (prolactinoma).

Pregnancy & Lactation: The safety of domperidone has not been proven, and it is therefore not recommended during pregnancy unless the anticipated therapeutic benefit justifies the potential risk. Domperidone may precipitate galactorrhoea and improve post-natal lactation. It is secreted in breast milk in very small quantities insufficient to be considered harmful.  

Storage:Store below 30°C, protected from light & moisture. Keep out of reach of children.
