Duphaston 10

Presentation: Each tablet contains 10 mg of dydrogesterone.

Indications: Duphaston 10 is indicated for the treatment of various conditions related to progesterone deficiency in women, including:

• Endometriosis
• Infertility due to luteal insufficiency
• Threatened miscarriage
• Habitual (recurrent) miscarriage
• Dysmenorrhea (painful menstruation)
• Irregular cycles
• Secondary amenorrhea (absence of menstruation)
• Premenstrual syndrome (PMS)
• Hormone Replacement Therapy (HRT) to counteract the effects of estrogen on the endometrium in postmenopausal women with an intact uterus.

Dosage and Administration: The dosage of Duphaston 10 varies depending on the condition being treated and should be strictly followed as prescribed by your doctor. The tablets are for oral administration and should be swallowed whole with a sufficient amount of liquid. The timing of the dose may also vary depending on the indication. For example:

• Endometriosis: Usually 10 mg two or three times daily, continuously from day 5 to day 25 of the cycle, or continuously.
• Infertility due to luteal insufficiency: Usually 10 mg daily from day 14 to day 25 of the cycle. Treatment should be continued for at least six consecutive cycles. It is advisable to continue the treatment during the first months of pregnancy at the same dosage as for habitual abortion.
• Threatened miscarriage: 40 mg as a single dose, then 10 mg every 8 hours.
• Habitual miscarriage: 10 mg twice daily until the 20th week of pregnancy, then gradually reduced.
• Dysmenorrhea: 10 mg twice daily from day 5 to day 25 of the cycle.
• Irregular cycles: 10 mg twice daily from day 11 to day 25 of the cycle.
• Secondary amenorrhea: Estrogen should be given once daily from day 1 to day 25 of the cycle, together with 10 mg dydrogesterone twice daily from day 11 to day 25 of the cycle.  
• PMS: Usually 10 mg twice daily from day 11 to day 25 of the cycle.
• HRT: When estrogen is given continuously, 10 mg dydrogesterone daily for the last 12 to 14 days of the estrogen cycle. When estrogen is given cyclically, 10 mg dydrogesterone daily for the last 12 to 14 days of each estrogen cycle.

Side Effects: Common side effects may include headache, nausea, and breast tenderness. Less common side effects can include migraine, dizziness, vomiting, allergic skin reactions (e.g., rash, itching, hives), weight increase, and mood changes. In rare cases, liver problems with jaundice, abdominal pain, or malaise have been reported. If you experience any persistent or troublesome side effects, you should inform your doctor.

Precautions: Duphaston 10 is contraindicated in patients with known hypersensitivity to dydrogesterone or any of the excipients, and in cases of known or suspected progestogen-dependent neoplasms (e.g., meningioma). Caution should be exercised in patients with a history of thromboembolic disorders (e.g., deep vein thrombosis, pulmonary embolism). If any signs of thromboembolic disorders occur, treatment should be discontinued. In case of breakthrough bleeding or spotting during HRT, endometrial hyperplasia should be excluded.

Storage: Store below 30°C in a dry place, protected from light. Keep out of reach of children.
