Nabupain (Nabumetone)

Indications Nabupain, a Non-Steroidal Anti-Inflammatory Drug (NSAID), is indicated for the relief of signs and symptoms of:

• Osteoarthritis
• Rheumatoid Arthritis

Pharmacology Nabumetone is a prodrug that exhibits anti-inflammatory, analgesic, and antipyretic properties.

It works by undergoing conversion in the liver to its active component, 6-methoxy-2-naphthylacetic acid (6MNA), which is a potent inhibitor of prostaglandin synthesis. Inhibition of prostaglandin synthesis is believed to cause its anti-inflammatory effects.

Dosage and Administration (Adults) The lowest effective dose should be used for the shortest duration.

• Initial Recommended Dose:1000 mg taken as a single dose (with or without food).
• Maximum Daily Dose Studied:2000 mg per day.
• Administration: Can be given in either a single or twice-daily dose.

Contraindications and Warnings

• Absolute Contraindications:
  • Hypersensitivity to Nabumetone or its excipients.
  • History of asthma, urticaria, or allergic reactions after taking aspirin or other NSAIDs.
  • Treatment of peri-operative pain in the setting of Coronary Artery Bypass Graft (CABG) surgery.
• Renal Toxicity: Use with caution in patients with impaired renal function, heart failure, or liver dysfunction. Discontinuation is usually followed by recovery.
• Pregnancy: Use during the third trimester is not recommended due to potential effects on the fetal cardiovascular system.

Side Effects Commonly reported side effects include:

• Gastrointestinal: Diarrhea, dyspepsia, abdominal pain, nausea, constipation.
• Central Nervous System: Dizziness, headache, fatigue, insomnia.
• Other: Pruritus (itching), rash, edema, tinnitus.

Interactions NSAIDs may reduce the antihypertensive effect of ACE-inhibitors.
