Protonil 40 

Presentation: Each enteric-coated tablet contains Pantoprazole Sodium Sesquihydrate INN equivalent to Pantoprazole 40 mg. Protonil is a brand name marketed by Renata PLC in Bangladesh. It is usually available in blister packs. The enteric coating helps to protect the medication from stomach acid, ensuring it is released in the intestine.

Indications: Protonil (Pantoprazole) is a proton pump inhibitor indicated for the treatment of acid-related disorders, including:

• Erosive Esophagitis associated with Gastroesophageal Reflux Disease (GERD).  
• Maintenance of healing of Erosive Esophagitis andreduction in relapse rates of daytime and nighttime heartburn symptomsin patients with healed EE.
• Treatment of pathological hypersecretory conditions including Zollinger-Ellison Syndrome.
• Peptic ulcer diseases (gastric and duodenal ulcers).
• Prevention of NSAID-induced ulcers.
• In combination with antibiotics for the eradication of Helicobacter pylori.

Dosage and Administration: Protonil 40 mg tablets should be swallowed whole, one hour before a meal, preferably in the morning. Do not chew or crush the tablets.

• Healing of Erosive Esophagitis: The usual adult dose is 40 mg once daily for up to 8 weeks. An additional 8-week course may be considered if healing is not complete.
• Maintenance of Healing of Erosive Esophagitis: The recommended adult dose is 40 mg once daily.
• Symptomatic GERD: The usual adult dose is 20 mg once daily for up to 4 weeks.
• Peptic Ulcer Disease: The usual adult dose is 40 mg once daily, usually for 2-4 weeks for duodenal ulcers and 4-8 weeks for gastric ulcers.
• Prevention of NSAID-induced ulcers: 20 mg once daily for patients requiring long-term NSAID treatment.
• Zollinger-Ellison Syndrome: The starting dose is usually 80 mg once daily, adjusted based on acid secretion.
• H. pylori eradication: 40 mg twice daily in combination with specific antibiotics for one week.

Side Effects: Pantoprazole is generally well-tolerated. Common side effects may include headache, diarrhea, nausea, abdominal pain, and flatulence. Less common side effects can include dizziness, dry mouth, and skin rash. Long-term use may be associated with an increased risk of bone fractures, low magnesium levels, and vitamin B12 deficiency in some individuals.

Precautions: Protonil (Pantoprazole) is contraindicated in patients with known hypersensitivity to pantoprazole or any other components of the formulation. Gastric malignancy should be excluded before initiating therapy. Caution should be exercised in patients with severe liver disease and during pregnancy and breastfeeding; consult your doctor before use. Pantoprazole can interact with several other medications, including some HIV medications, antifungals, and anticoagulants.

Storage: Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
