Prugen 2 

Presentation: Prugen 2, available in Bangladesh, contains Prucalopride Succinate equivalent to Prucalopride 2 mg. It is presented as film-coated tablets for oral administration and is usually available in blister packs. 

Indications: Prugen (Prucalopride) is indicated for the symptomatic treatment of chronic constipation in adults in whom laxatives provide insufficient relief.

Dosage and Administration: The recommended oral dose of Prugen for adults is 2 mg once daily, taken with or without food, at any time of the day. The physician may initiate treatment at 1 mg once daily in the elderly (over 65 years) and, if needed, increase to 2 mg once daily. For patients with severe renal impairment (GFR < 30 ml/min/1.73 m²), the recommended starting dose is 1 mg once daily, which may be increased to 2 mg once daily if needed and if the 1 mg dose is well-tolerated.

The efficacy of Prucalopride may be enhanced if taken at the same time each day. The duration of treatment should be determined by the physician based on the patient's response and the need for continued relief.

Side Effects: The most commonly reported side effects with Prugen (Prucalopride) are headache, nausea, diarrhea, and abdominal pain. These side effects are usually mild to moderate and transient, occurring mainly at the start of therapy and subsiding with continued treatment. Other common side effects may include vomiting, dyspepsia, flatulence, and fatigue.

Less common side effects may include dizziness, palpitations, pollakiuria (frequent urination), and rash.

Precautions: Prugen (Prucalopride) is contraindicated in patients with known hypersensitivity to prucalopride or any of the excipients, severe renal impairment requiring dialysis, intestinal perforation or obstruction due to structural or functional disorder, obstructive ileus, severe inflammatory conditions of the intestine such as Crohn's disease, ulcerative colitis, and toxic megacolon/megarectum.

Caution should be exercised in patients with a history of cardiac arrhythmias or ischemic cardiovascular disease. If severe or persistent diarrhea occurs, the physician should be consulted, and caution should be exercised in patients who are also taking diuretics as this may increase the risk of hypokalemia.

Use in Pregnancy and Breastfeeding: There are limited data on the use of Prucalopride in pregnant women. Prugen is not recommended during pregnancy unless the potential benefit outweighs the potential risk to the fetus. Women of childbearing potentialshould use effective contraception during2treatment. Prucalopride is excreted in human milk. Breastfeeding is not recommended during treatment with Prugen. A decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.3  

Storage: Store below 30°C, protected from light and moisture. Keep out of reach of children.
