Indications: Prulax (Prucalopride) is a selective 5-HT4 receptor agonist indicated for the treatment of chronic constipation in adults when laxatives have failed to provide adequate relief.
Dosage and Administration: The recommended dose is 1 mg once daily, taken orally at any time of the day, with or without food. Based on individual patient response and tolerability, the dose may be increased to 2 mg once daily. Caution is advised when increasing the dose in elderly patients.
Side Effects:Common side effects may include headache, nausea, diarrhea, and abdominal pain. These side effects are usually mildto moderate and transient, occurring mainly at the start of therapy. Other possible side effects include vomiting, dyspepsia, dizziness, and fatigue.
Precautions: Prulax (Prucalopride) is contraindicated in patients with known hypersensitivity to prucalopride or any of the excipients, intestinal obstruction or perforation due to mechanical or obstructive ileus, severe inflammatory conditions of the intestine such as Crohn's disease and ulcerative colitis, and toxic megacolon. Caution should be exercised in patients with severe renal impairment (reduce starting dose to 1 mg once daily), severe hepatic impairment, and in patients with a history of cardiac arrhythmias or ischemic cardiovascular disease.
Use in Pregnancy and Lactation: There are limited data on the use of prucalopride in pregnant women. Animal studies do not indicate direct or indirect harmful effects with respect to reproductive toxicity. Prulaxshould be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Prucalopride is excreted in human milk. However, at therapeutic doses, no effects on breastfed newborns/infants are anticipated. As a precautionary measure, it is preferable to avoid the use of Prulax during breastfeeding.
