Seasonix (Levocetirizine): A Non-Sedating Antihistamine

Seasonix contains Levocetirizine Dihydrochloride, the active (Levo) isomer of cetirizine. It is a highly effective, well-tolerated, non-sedating antihistamine with potent antiallergic properties. It is indicated for treating symptoms associated with perennial allergic rhinitis, seasonal allergic rhinitis, and chronic idiopathic urticaria. It belongs to the therapeutic class of Non-sedating antihistamines.


How Seasonix Works (Pharmacology)

Levocetirizine's mechanism of action involves:

  • H1 Receptor Affinity: Levocetirizine is an antihistamine that has a high and selective affinity for H1 receptors. It has been shown to have a two-fold higher affinity for these receptors than its parent compound, cetirizine.
  • Antiallergic Properties: By binding to and blocking H1 receptors, Levocetirizine prevents histamine from activating these receptors. Histamine is a key mediator of allergic and hypersensitivity reactions. This action helps to alleviate a wide range of allergic symptoms such as sneezing, rhinorrhea (runny nose), and itching.
  • Rapid and Long-Acting: The drug has a rapid onset of action and a long-lasting effect, which allows for convenient once-a-day administration.
  • Non-Sedating: As a non-sedating antihistamine, it is less likely to cause drowsiness or other CNS effects because it does not readily cross the blood-brain barrier.

Key Indications & Benefits

Seasonix is indicated for the treatment of symptoms associated with allergic conditions such as:

  • Perennial allergic rhinitis.
  • Seasonal allergic rhinitis.
  • Chronic idiopathic urticaria (hives).

Dosage & Administration

Dosage is based on age and varies for patients with renal impairment. Always consult a registered physician for specific dosage instructions.

  • Adults and children 12 years of age and older: The recommended dose is 5 mg once daily.
  • Children 6 to 11 years of age: The recommended dose is 2.5 mg (half a tablet or 1 teaspoon oral solution) once daily.
  • Children 6 months to 5 years of age: The recommended dose is 1.25 mg (half a teaspoon oral solution) once daily.

Use in Specific Populations:

  • Geriatric use: Insufficient data from clinical studies to determine if elderly patients respond differently than younger patients.
  • Hepatic impairment: No dosage adjustment is required for patients with solely hepatic impairment.
  • Renal impairment:
    • Mild (creatinine clearance: 50-80 ml/min): 2.5 mg once daily.
    • Moderate (creatinine clearance: 30-50 ml/min): 2.5 mg once every other day.
    • Severe (creatinine clearance: 10-30 ml/min): 2.5 mg twice weekly (once every 3-4 days).
    • End-stage renal disease (creatinine clearance: <10 ml/min) and patients on hemodialysis: Should not receive Levocetirizine.

Contraindications:

  • Hypersensitivity to levocetirizine, cetirizine, or its parent compound, hydroxyzine.
  • Severe renal impairment (creatinine clearance <10 ml/min).

Side Effects:

  • Levocetirizine is generally well tolerated.
  • Rarely reported side effects: Headache, dry mouth, fatigue, and skin rash.

Pregnancy & Lactation:

  • Pregnancy: Classified as Pregnancy Category B. There are no adequate and well-controlled studies in pregnant women. Use during pregnancy only if clearly needed.
  • Lactation: Levocetirizine is excreted in breast milk. It is not recommended for use by nursing mothers.

Precautions & Warnings:

  • Use with caution in epileptic patients and patients at risk of convulsions.
  • Exercise caution in patients with renal impairment.
  • Use during pregnancy and lactation should be carefully considered and discussed with a physician.

Overdose Effects

  • No clinically relevant adverse events have been reported in cases of overdose.
  • Treatment: In the event of an overdose, symptomatic and supportive treatment is recommended.
  • Hemodialysis: Levocetirizine is not effectively removed by hemodialysis.

Store at a temperature not exceeding 30°C in a dry place. Protect from light. Keep out of the reach of children.
