Indications

Sinamin (Chlorpheniramine Maleate) is a potent sedating antihistamine indicated for the treatment of:

  • Urticaria (hives)
  • Sensitivity reactions
  • Angioneurotic edema
  • Vasomotor rhinitis
  • Cough and common cold
  • Motion sickness
  • Other allergic conditions

 

Pharmacology

Chlorpheniramine Maleate is an alkylamine antihistamine that acts as a competitive antagonist of the histamine H1-receptor. By blocking these receptors, it prevents the effects of histamine, which are responsible for a wide range of allergic symptoms. It generally causes less sedation than some other antihistamines like promethazine.

 

Dosage & Administration

  • Adults: 4 mg every 4-6 hours, with a maximum daily dose of 24 mg.
  • Children:
    • 6-12 years: 2 mg every 4-6 hours, with a maximum daily dose of 12 mg.
    • 2-5 years: 1 mg every 4-6 hours, with a maximum daily dose of 6 mg.
    • 1-2 years: 1 mg twice daily.
    • Below 1 year: Use is not recommended.

 

Interaction

Chlorpheniramine Maleate has been reported to be incompatible with calcium chloride, kanamycin sulfate, noradrenaline acid tartrate, pentobarbital sodium, and meglumine adipiodone.

 

Contraindications

Sinamin is contraindicated in patients with a known hypersensitivity to the drug, as well as in newborn or premature infants.

 

Side Effects

Sinamin is generally well-tolerated, but common side effects include drowsiness, dizziness, muscular weakness, and gastrointestinal upset.

 

Pregnancy & Lactation

This drug should not be used by breastfeeding mothers or during pregnancy, especially in the first trimester.

 

Precautions & Warnings

  • Use with caution in patients with glaucoma and prostatic hypertrophy.
  • Due to the potential for drowsiness, caution should be taken when driving vehicles or operating heavy machinery.

 

Overdose Effects

Overdose may cause central nervous system (CNS) depression (e.g., sedation, cardiovascular collapse) or CNS stimulation (e.g., insomnia, tremors, convulsions). Other symptoms may include blurred vision, dizziness, hypotension, and atropine-like effects such as dry mouth and hyperthermia, which are more common in children.

 

Therapeutic Class

Sinamin is a Sedating Anti-histamine.

 

Storage Conditions

Store in a cool and dry place below 25°C, protected from light. Keep out of the reach of children.
