Presentation:Each film-coated tablet contains Prucalopride Succinate INN equivalent to Prucalopride 1 mg.Sinapride 1 is a product of Ibn Sina Pharmaceutical Industry Ltd. in Bangladesh and is available in blister packs.
Indications:Sinapride (Prucalopride) is indicated for the symptomatic treatment of chronic constipation in adults in whom laxatives fail to provide adequate relief.
Dosage and Administration: The recommended dose for adults is 2 mg once daily with or without food, at any time of the day.However, for older people, the starting dose is 1 mg once daily, which can be increased to 2 mg once daily if needed. For patients with severe renal impairment (GFR < 30 ml/min/1.73 m²), the recommended dose is 1 mg once daily.Prucalopride is not recommended for use in children and adolescents younger than 18 years. Due to Prucalopride's specific action of stimulating propulsive motility, exceeding a daily dose of 2 mg is not expected to increase efficacy.
Side Effects:The most frequently reported adverse reactions associated with Sinapride (Prucalopride) therapy are headache and gastrointestinal symptoms such as abdominal pain, nausea, and diarrhoea.These reactions occur predominantly at the start of therapy and usually disappear within a few days with continued treatment.Other adverse reactions have been reported occasionally,and the majority of events were mild to moderate in intensity.
Precautions:Sinapride (Prucalopride) is contraindicated in individuals with hypersensitivity to prucalopride or any of the excipients and in patients with renal impairment requiring dialysis.Caution should be exercised when prescribing Prucalopride to patients with severe hepatic impairment due to limited data. In case of severe diarrhoea, the efficacy of oral contraceptives may be reduced, and using an additional contraceptive method is recommended.The tablets contain lactose, so patients with rare hereditary problems of galactose intolerance, the Lapp lactase deficiency, or glucose-galactose malabsorption should not take this medicine.
Use in Pregnancy and Lactation: Prucalopride is not recommended during pregnancy, and women of childbearing potential should use effective contraception during treatment.In the absence of human data, it is not recommended to use Prucalopride during breastfeeding.
Storage:Store at room temperature, below 30°C.Keep out of the reach of children.
