Sizonil 5 Tablet contains Trifluoperazine Hydrochloride equivalent to 5 mg of Trifluoperazine. It is manufactured by Healthcare Pharmaceuticals Ltd.

Indications: Sizonil is used for:

  • Management of psychotic disorders such as schizophrenia.
  • Short-term management of severe anxiety.
  • Treatment of nausea and vomiting.

Pharmacology: Trifluoperazine is a phenothiazine antipsychotic. It works by blocking dopamine receptors in the brain, which helps to reduce psychotic symptoms and anxiety. It also has antiemetic properties by blocking dopamine receptors in the chemoreceptor trigger zone.

Dosage and Administration: The dosage varies depending on the condition and the patient's response. It is usually taken orally, with or without food. Some general guidelines:

  • Psychotic disorders (Schizophrenia):
    • Adults and children over 12 years: Usually start with 2-5 mg twice daily, gradually increased by the doctor.
    • Children (6-12 years): Dosage adjusted by weight and severity, starting at 1 mg twice daily, gradually increased.
    • Elderly: Lower initial dose, adjusted by the doctor.
  • Short-term management of severe anxiety:
    • Adults and children over 12 years: 1-2 mg twice daily, increased if needed up to 6 mg daily.
    • Children (3-5 years): 1 mg daily.
    • Children (6-12 years): Up to 4 mg daily in divided doses.
    • Elderly: Lower initial dose, adjusted by the doctor.
  • Antiemetic:
    • Adults: 2-4 mg daily in divided doses, maximum 6 mg daily.
    • Children (3-5 years): Up to 1 mg daily.
    • Children (6-12 years): Up to 4 mg daily.

Side Effects: Common side effects may include:

  • Restlessness
  • Muscle spasms or stiffness
  • Parkinsonism-like symptoms (tremors, slow movement)
  • Drowsiness
  • Dizziness
  • Fatigue
  • Blurred vision
  • Seizures
  • Dry mouth
  • Loss of appetite

Less common but potentially serious side effects can occur. It is important to report any unusual or severe symptoms to your doctor immediately.

Precautions:

  • Use with caution in elderly patients, and start with a lower dose.
  • Use with caution in patients with cardiovascular disease.
  • May cause drowsiness; avoid driving or operating machinery until you know how it affects you.
  • Avoid alcohol consumption as it can increase sedation.
  • Inform your doctor about any pre-existing medical conditions, especially epilepsy, glaucoma, enlarged prostate, or heart problems.
  • Be aware of potential weight gain, and maintain a healthy diet and exercise regularly.
  • May increase blood sugar and cholesterol levels; regular monitoring may be needed.
  • May make you more sensitive to temperature changes.

Contraindications: Sizonil is contraindicated in patients with:

  • Comatose conditions.
  • Existing blood disorders (dyscrasias).
  • Known liver damage.
  • Hypersensitivity to trifluoperazine or related compounds.

Drug Interactions: Sizonil can interact with various medications. Inform your doctor about all the medicines you are currently taking, including:

  • Oral anticoagulants (may reduce their effect).
  • Propranolol (may increase levels of both drugs).
  • Guanethidine and related antihypertensive compounds (may counteract their effects).
  • CNS depressants like alcohol, hypnotics, and anticonvulsants (may increase sedation).

Pregnancy and Lactation: The safety of trifluoperazine during pregnancy has not been fully established and is generally not recommended unless the potential benefits outweigh the risks. It may pass into breast milk and harm the baby; breastfeeding is generally not recommended during treatment.

Storage: Store below 30°C, protected from light and moisture. Keep out of reach of children.
