Supraphen, Ophthalmic Solution - (0.50%) contains Chloramphenicol 0.5%.It is manufactured by Gaco Pharmaceuticals Ltd.  

Indications:

Supraphen eye drops are indicated for the treatment of ocular infections involving the conjunctiva and/or cornea caused by susceptible organisms, including:

  • Staphylococcus aureus
  • Streptococcus pneumoniae
  • Escherichia coli
  • Haemophilus influenzae
  • Klebsiella/Enterobacter spp.
  • Moraxella lacunata
  • Neisseria species

Pharmacology:

Chloramphenicol is a broad-spectrum bacteriostatic antibiotic. It inhibits bacterial protein synthesis by interfering with the transfer of activated amino acids from soluble RNA to ribosomes. 

Dosage and Administration:

  • Adults and Children: Instill 1 or 2 drops in the conjunctival sac 4 to 6 times per day for the first 72 hours, and then every 4 hours thereafter. 
  • Treatment should be continued for approximately 7 days but should not exceed 3 weeks without re-evaluation by a physician.

Contraindications:

Supraphen is contraindicated in individuals with a history of hypersensitivity to Chloramphenicol or any other ingredients in the preparation. 

Side Effects:

Systemic adverse reactions are not commonly observed with short-term topical use. The most frequently reported side effects include:

  • Burning
  • Stinging 
  • Conjunctival hyperemia (redness of the eye) 
  • Allergic or inflammatory reactions
  • Vesicular and maculopapular dermatitis (skin rashes) 

Precautions and Warnings:

  • Supraphen ophthalmic solution should never be given for minor infections or for prophylaxis. 
  • Repeated courses and prolonged treatment should be avoided. 
  • Blood dyscrasias (serious blood disorders) may occur after prolonged ophthalmic use, although this is rare. 
  • Avoid touching the dropper tip to any surface, including the eye, to prevent contamination. 
  • Do not wear contact lenses until the infection clears up. 

Pregnancy and Lactation:

Safety for use during pregnancy and lactation has not been established. Use only when considered essential by a physician.

Storage Conditions:

Store in a cool place (between 2°C and 8°C) and protect from light. Do not use after 30 days of first opening. Keep out of reach of children.
