Vonosil 20 

Presentation: Each film-coated tablet of Vonosil 20 contains Vonoprazan Fumarate INN equivalent to Vonoprazan 20 mg.

Indications: Vonosil (Vonoprazan) is a potassium-competitive acid blocker (P-CAB) indicated for:

  • Treatment of gastric ulcer.
  • Treatment of duodenal ulcer.
  • Treatment of reflux esophagitis (erosive esophagitis).
  • Prevention of recurrence of gastric ulcer or duodenal ulcer during low-dose aspirin administration.  
  • Prevention of recurrence of gastric ulcer or duodenal ulcer during NSAIDs administration.  
  • Adjunctto Helicobacter pylorieradicationin the following conditions: gastric ulcer, duodenal ulcer, gastric mucosa-associated lymphatic tissue (MALT) lymphoma, idiopathic thrombocytopenic purpura, the stomach after endoscopic resection of early-stage gastric cancer,or Helicobacter pylorigastritis.  

Dosage and Administration: Vonosil 20 tablets should be taken orally without regard to food or timing of food.

  • Gastric ulcer: The usual adult dose is 20 mg once daily for up to 8 weeks.
  • Duodenal ulcer: The usual adult dose is 20 mg once daily for up to 6 weeks.
  • Reflux esophagitis: The usual adult dose is 20 mg once daily for up to 4 weeks. If healing is incomplete, treatment may be extended up to 8 weeks. For maintenance therapy of healed erosive esophagitis, the usual dose is 10 mg once daily, which may be increased to 20 mg once daily if efficacy is inadequate.
  • Prevention of recurrence of gastric or duodenal ulcer during NSAIDs administration: The dose is 10 mg once daily.
  • Prevention of recurrence of gastric or duodenal ulcer during low-dose aspirin administration: The dose is 10 mg once daily.
  • Helicobacter pylori eradication: The usual regimen is Vonoprazan 20 mg, Amoxicillin 750 mg, and Clarithromycin 200 mg, all taken twice daily for 7 days. The dose of Clarithromycin may be increased up to 400 mg twice daily if clinically warranted.

The dosage and duration of treatment should be determined by a physician based on the individual patient's condition.

Side Effects: The most common side effects reported with Vonosil (Vonoprazan) include:

  • Diarrhoea
  • Constipation
  • Nausea
  • Abdominal distension
  • Increased liver enzymes (ALT, AST, gamma-glutamyl transferase)
  • Rash

Other side effects may include drug hypersensitivity reactions (such as urticaria and anaphylactic shock), jaundice, and abnormal liver function tests.

Precautions: Vonosil (Vonoprazan) is contraindicated in patients with hypersensitivity to Vonoprazan or any excipients in the product, and in patients receiving atazanavir sulphate, nelfinavir, or rilpivirine hydrochloride.

Caution should be exercised in patients with liver function abnormalities. Gastric malignancy should be excluded before starting treatment. Vonoprazan may interact with other drugs; inform your doctor about all medications you are taking.

Use in Pregnancy and Lactation: Vonosil should be used during pregnancy only if the expected therapeutic benefit outweighs the possible risk to the foetus. It is advisable to avoid administration to nursing mothers; if deemed necessary, breastfeeding should be discontinued during treatment.

Storage: Store below 30°C, in a cool and dry place, protected from light. Keep out of the reach of children.
