Vortiox 10 Tablet typically contains Vortioxetine Hydrobromide equivalent to 10 mg of Vortioxetine.

Indications: Vortioxetine is indicated for the treatment of Major Depressive Disorder (MDD) in adults. 

Pharmacology:The mechanismof action of vortioxetine is not fully understood. It is thought to be related to its enhancement of serotonergic activity in the central nervous system through the inhibition of serotonin (5-HT) reuptake. Additionally, vortioxetine has several other activities, including:  

• 5-HT1A receptor agonism
• 5-HT1B receptor partial agonism
• 5-HT1D receptor antagonism
• 5-HT3 receptor antagonism
• 5-HT7 receptor antagonism

This multimodal activity is thought to contribute to its antidepressant effects and potential pro-cognitive effects.

Dosage and Administration: The recommended starting dose is 10 mgadministered orally once daily, without regard to meals. The dosage may be increased to 20 mg/day as tolerated. A dose decrease down to 5 mg/day may be considered for patients who do not tolerate higher doses. The lowest effective dose of 5 mg/dayshould be used as the starting dose for elderly patients (>65 years of age).  

Contraindications: Vortioxetine is contraindicated in patients with:

• Known hypersensitivity to vortioxetine or any of the excipients.
• Concomitant use of Monoamine Oxidase Inhibitors (MAOIs). There must be at least 14 days between discontinuation of an MAOI and initiation of vortioxetine, and at least 21 days after stopping vortioxetine before starting an MAOI.

Precautions:

• Suicidal Thoughts and Behaviors: Monitor patients for clinical worsening and suicidal thoughts and behaviors, especially during the initial months of therapy and when the dose is changed.
• Serotonin Syndrome: Be aware of the potential for serotonin syndrome, especially with concomitant use of other serotonergic drugs.
• Increased Risk of Bleeding: Vortioxetine may increase the risk of bleeding events.
• Activation of Mania/Hypomania: Use with caution in patients with a history of mania.
• Angle-Closure Glaucoma: May trigger an angle-closure attack in patients with anatomically narrow angles.
• Hyponatremia: Low sodium levels in the blood have been reported.

Side Effects: Common side effects may include:

• Nausea
• Headache
• Dry mouth
• Dizziness
• Constipation
• Vomiting
• Abnormal dreams
• Diarrhea
• Flatulence
• Itching

Less common but potentially serious side effects can occur. It is important to report any unusual or severe symptoms to your doctor immediately.

Drug Interactions:

• MAOIs: Contraindicated due to the risk of serotonin syndrome.
• Other Serotonergic Drugs: Use with caution due to the increased risk of serotonin syndrome.
• CYP2D6 Inhibitors: May increase vortioxetine plasma concentrations; a dose reduction of vortioxetine may be necessary.
• CYP Inducers: May decrease vortioxetine plasma concentrations; a dose increase of vortioxetine may be considered.

Storage: Store as directed on the product packaging, usually at room temperature, away from light and moisture. Keep out of reach of children.
